Optional, defined, and never a prerequisite.
The paper is already sustainable from documentary evidence. If pursued later, the defensible question is narrow: does the documented direct paternal line share a recent common male ancestor with a documented York/Chanceford Staley or Stehli candidate line?
A carefully documented Y-DNA comparison could test compatibility with a paternal branch. It could not, by itself, prove that the 1793 Berkeley testator was a particular York man. Any testing, upload, match contact or publication requires a separate private decision.
